Nicotinamide mononucleotide adenylyl transferase 2 (NMNAT2) is a key neuronal maintenance factor and provides potent neuroprotection in many preclinical models of neurological disorders. NMNAT2 is significantly reduced in Alzheimer’s, Huntington’s and Parkinson’s diseases.
Here, we have developed a discovery-based mesoscale (MSD) screening platform to quantify endogenous NMNAT2 in cortical neurons. The high sensitivity and large dynamic range of this NMNAT2-MSD platform allowed us to screen the Sigma LOPAC library composed of 1280 compounds. This library had a success rate of 2.89%, with 24 positive and 13 negative NMNAT2 modulators identified. Western analysis was conducted to validate and determine the dose dependence of the identified modulators.
Caffeine, an identified positive modulator of NMNAT2, when administered systemically restores the expression of NMNAT2 in rTg4510 tauopathy mice to normal levels. We confirmed in a cell culture model that four selected positive modulators exerted NMNAT2-specific neuroprotection against vincristine-induced cell death while four selected NMNAT2 negative modulators reduced neuronal viability in an NMNAT2-dependent manner. It is predicted that many of the positive NMNAT2 modulators identified will increase the concentration of cAMP, suggesting that neuronal levels of NMNAT2 are tightly regulated by cAMP signaling. Taken together, our results indicate that the NMNAT2-MSD platform provides a sensitive phenotypic screen for detecting NMNAT2 in neurons.
A rapid innovative method for the analysis of 10 organophosphate pesticide residues in wheat by HS-SPME-GC-FPD / MSD.
The rapid detection of pesticide residues in wheat has become a top priority in food safety. Solid phase microextraction (HS-SPME) in the solvent-free headspace was evaluated for the rapid detection of organophosphate pesticide (OPP) residues in high sensitivity wheat. Individual wheat samples (1.7g), enriched with 10 OPP, were placed in a sealed 4ml amber glass vial and heated at 60 ° C for 45 minutes. During this time, the OPP residues were extracted with a 50 μm / 30 μm polydimethylsiloxane (PDMS) fiber of divinylbenzene (DVB) / carboxene (CAR) / plasma desorption mass spectroscopy (PDMS) at the headspace. above the sample.
The fiber was then removed and injected into the orifice of injection of GC at 250 ° C for the desorption of the extracted chemicals.
